Cell-cycle Dependent Desquamation of Heparan Sulfate from the Cell Surface

چکیده
The term heparin sulfate denotes mucopolysaccharides with a structure similar to heparin but which lack significant anticoagulant potency (1) . The term also connotes a continuum or family of N-sulfated mucopolysaccharides of which the more highly sulfated varieties are called heparins . It appears likely that all cultured mammalian cells synthesize heparan sulfate (2, 3) and that this material becomes incorporated into a metabolically active fraction of the cell surface (4) . The heparan sulfate of the cell surface is readily removed by gentle trypsin treatment under conditions where no irreversible cell damage occurs ; in the case of Chinese hamster cells (line CHO), trypsin treatment removes heparan sulfate and a melange of glycopeptides from cell-surface glycoproteins (4) . These two categories of cell-surface materials have independent metabolism. The ubiquity of heparan sulfate in cultured mammalian cells suggests some general role in cellular metabclism. We report here results concerning cellcycle related fluctuations in the amount present on the cell surface as well as variations in labeling rate from supplied glucosamine-6 3H . Our results encourage speculations as to the role of a generally occurring cell-surface component in the regulation of exposure of components with specific functions for a particular cell type .

Correlation between cell substrate attachment in vitro and cell surface heparan sulfate affinity for fibronectin and collagen
Heparan sulfate glycosaminoglycan, isolated from the cell surface of nonadhering murine myeloma cells (P3X63-Ag8653), does not bind to plasma fibronectin, but binds partially to collagen type I, as assayed by affinity chromatography with proteins immobilized on cyanogen bromide-activated Sepharose 4B. Identical results were obtained when myeloma heparan sulfate was cochromatographed, on the sam...

Background and Objective: Hybrid cells are made from fusion of two or more somatic cells. After formation chromosomes are located in one membrane . . So nucleic condition of each fused cell has changes and two genomes and chromosomes interact with each other. Locating the genes in new nucleic and cytoplasmic condition and great chromosomal rearrangement in these new formed cells can affect th...

Cell surface heparan sulfate proteoglycans facilitate uptake of growth-promoting polyamines (Belting, M., Borsig, L., Fuster, M. M., Brown, J. R., Persson, L., Fransson, L.-A., and Esko, J. D. (2002) Proc. Natl. Acad. Sci. U. S. A. 99, 371-376). Increased polyamine uptake correlates with an increased number of positively charged N-unsubstituted glucosamine units in the otherwise polyanionic hep...
متن کاملHeparan sulfate proteoglycan on leukemic cells is primarily involved in integrin triggering and its mediated adhesion to endothelial cells
Leukocyte migration from circulation into tissue depends on leukocyte integrin-mediated adhesion to endothelium, but integrins cannot function until activated. However, it remains to be understood how tumor cells adhere to endothelium and infiltrate into underlying tissue. We studied mechanisms of extravasation of leukemic cells using adult T cell leukemia (ATL) cells and report the following n...
متن کاملHeparan sulfate regulates ephrin-A3/EphA receptor signaling.
Increasing evidence indicates that many signaling pathways involve not only ligands and receptors but also various types of coreceptors and matrix components as additional layers of regulation. Signaling by Eph receptors and their ephrin ligands plays a key role in a variety of biological processes, such as axon guidance and topographic map formation, synaptic plasticity, angiogenesis, and canc...
